Overview

InviTaq Hot Start DNA Polymerase is supplied in an inactive state that has no polymerase activity at ambient temperatures. This enzyme is recommended for all PCR and real-time PCR reactions. The 5’-3’ DNA exonuclease activity has a broad thermal range (50-74°C). The thermal activation prevents the extension of nonspecifically annealed primers and primer dimers formed at low temperatures during PCR setup. The polymerase contains monoclonal antibodies which block polymerase activity prior to the onset of thermal cycling. The enzyme requires no prolonged heating or denaturing step. The antibodies are inactivated during an initial denaturation step of 5 minute at 95°C which can be incorporated into any existing thermal-cycler program. The InviTaq Hot Start DNA Polymerase lacks 3’-5’ exonuclease activity.

Supplied Reaction Buffers:

1) 10x Reaction Buffer

250 mM Tris-HCl pH 8.3 (at 25 °C), 500 mM KCl
2) Mg2+Solution
50 mM MgCl2 (recommended final concentration: 1 – 6 mM)

Workflow

The Hot Start DNA Polymerase is activated in the 5-minute incubation at 95°C before PCR cycling.
